The Ruff Ryders namesake also referred to a loose-knit hip-hop collective composed of the core signees of the label. Ruff Ryders and its main studio Powerhouse Studios are headquartered at 33 South Broadway in Yonkers, New York.

Mason Durell Betha, known professionally as Mase, is an American rapper. Best known for his work with Sean "Puff Daddy" Combs' Bad Boy Records, he signed with the label in 1996 and quickly found mainstream recognition as Combs' hype man. Michael Lawrence Tyler, better known by his stage name Mystikal, is an American former rapper and actor from New Orleans, Louisiana. He is noted for his brash and explosive vocal delivery, characterized by a Southern rasp. He signed with the New Orleans-based record label Big Boy Records to release his self-titled debut studio album (1994), which was re-released by Jive Records the following year as his major label debut, Mind of Mystikal (1995). He then parted ways with the former label in favor of its local competitor, Master P's No Limit Records, through which he released his second and third albums, Unpredictable (1997) and Ghetto Fabulous (1998), and Goodfellas (1999) as a member of its flagship group, 504 Boyz.

Barry Adrian Reese, better known by his stage name Cassidy, is an American rapper. He first garnered attention for his freestyles and competitions as a battle rapper in the late 1990s, and formed the Philadelphia-based hip hop collective Larsiny by the end of the decade. American record producer Swizz Beatz discovered the group in 2000 and signed Cassidy to his newly-established Full Surface Records, in a joint venture with J Records. Timbaland & Magoo were an American hip-hop duo composed of Virginia natives Timothy "Timbaland" Mosley and Melvin "Magoo" Barcliff (1973–2023). LaRon Louis James Sr., better known by his stage name Juelz Santana, is an American rapper. A member of now-defunct hip-hop group the Diplomats, he is best known for his appearances on Cam'ron's 2002 singles "Oh Boy" and "Hey Ma", both of which peaked within the top five of the Billboard Hot 100. He later guest appeared on Chris Brown's 2005 single, "Run It!", which peaked atop the chart for five weeks and received triple platinum certification by the Recording Industry Association of America (RIAA).
