The Ruff Ryders namesake also referred to a loose-knit hip-hop collective composed of the core signees of the label. Ruff Ryders and its main studio Powerhouse Studios are headquartered at 33 South Broadway in Yonkers, New York.

John David Jackson, known professionally as Fabolous, is an American rapper. Raised in Brooklyn, he first gained recognition for his ability upon performing live on DJ Clue's Hot 97 radio show. Jackson then signed to Clue's record label Desert Storm Records, in a joint venture with Elektra Records. He rose to further prominence with his debut studio album Ghetto Fabolous (2001), which spawned the hit singles "Can't Deny It" and "Young'n ." LaRon Louis James Sr., better known by his stage name Juelz Santana, is an American rapper. A member of now-defunct hip-hop group the Diplomats, he is best known for his appearances on Cam'ron's 2002 singles "Oh Boy" and "Hey Ma", both of which peaked within the top five of the Billboard Hot 100. He later guest appeared on Chris Brown's 2005 single, "Run It!", which peaked atop the chart for five weeks and received triple platinum certification by the Recording Industry Association of America (RIAA). Patrick Owen Baril, known professionally as Statik Selektah, is an American DJ and record producer from Boston, Massachusetts. He is also the founder of Showoff Records. In 2010, he formed the hip hop group 1982, alongside American rapper Termanology. He has resided in New York City since 2004.

Jay Wayne Jenkins, known by his stage name Jeezy, is an American rapper. He is credited, alongside fellow Georgia-based rappers T.I. and Gucci Mane, with pioneering the hip hop subgenre trap music for mainstream audiences. Jenkins began his career in 2001, releasing two independent albums until signing with Def Jam Recordings to release his major label debut, Let's Get It: Thug Motivation 101 (2005). Shante Scott Franklin, better known by his stage name Currensy, is an American rapper. Born and raised in New Orleans, Louisiana, he signed with the local record labels No Limit Records, Cash Money Records and Young Money Entertainment in the mid-2000s, but parted ways with both without any major releases. He signed with DD172 to release four albums before signing with Warner Records to release his fifth album and major label debut, Weekend at Burnie's (2011). In February of that year, he founded the record label Jet Life Recordings.

David Darnell Brown, better known by his stage name Young Buck, is an American rapper. He was affiliated with Birdman's Cash Money Records in 1997, formed the hip hop collective UTP with Juvenile and Soulja Slim in 2000, and joined 50 Cent's group, G-Unit by 2003. In 2003, he signed with 50 Cent's G-Unit Records, an imprint of Interscope Records to release his debut studio album Straight Outta Cashville (2004) and its follow-up Buck the World (2007), both of which peaked at number three on the Billboard 200 and were met with critical praise.

Mobb Deep were an American hip-hop duo formed in Queens, New York, in 1990. Consisting of rappers/songwriters/record producers Prodigy and Havoc, they are considered to be among the most influential artists in East Coast hip-hop. Mobb Deep are one of the most successful rap duos of all time, having sold over three million records. Karim Kharbouch, known professionally as French Montana, is a Moroccan and American rapper. Born and raised in Morocco, he immigrated to New York City with his family at the age of 13 and began his career as a battle rapper in the early 2000s — under the name Young French. He first gained recognition hosting the locally-tailored DVD series Cocaine City during the 2000s, which centered around interviews of hip hop figures. He pursued a recording career while doing so, releasing several underground projects before signing with Sean Combs's Bad Boy Records and Rick Ross's Maybach Music Group, in a triple-joint venture with Interscope Records in 2012.

The Beatnuts is an American hip hop group and production duo from New York City. Its current members are JuJu and Psycho Les. JuJu is a Dominican American from Corona and Psycho Les is a Colombian American from Jackson Heights, Queens. Although only peripheral members, they are routinely acknowledged by Q-Tip as being members of Native Tongues. The Beatnuts were originally a trio before Fashion, now known as Al' Tariq, left the group to start a solo career. V.I.C. was also a member of The Beatnuts' production team for a while.

Terrence LeVarr Thornton, better known by his stage name Pusha T, is an American rapper. He rose to prominence as one half of the Virginia-based hip hop duo Clipse, which he formed with his older brother Malice. Established in 1994, the duo were discovered by Pharrell Williams and signed with his record label Star Trak Entertainment, an imprint of Arista Records, in 2001. They initially released three studio albums—Lord Willin' (2002), Hell Hath No Fury (2006) and Til the Casket Drops (2009)—to mild commercial success; the former spawned the Billboard Hot 100-top 40 singles "Grindin'" and "When the Last Time", for which they became best known.

He first garnered attention for his freestyles and competitions as a battle rapper in the late 1990s, and formed the Philadelphia-based hip hop collective Larsiny by the end of the decade. American record producer Swizz Beatz discovered the group in 2000 and signed Cassidy to his newly-established Full Surface Records, in a joint venture with J Records. He adopted a commercially-oriented approach for his 2003 debut single "Hotel" and its follow-up, "Get No Better" ; the former peaked at number four on the Billboard Hot 100 while both preceded the release of his debut studio album, Split Personality (2004).
